About Brain Barriers Research This review aims to provide an up to date and comprehensive guideline for researchers in diverse fields, offering perspectives on further development of brain targeted drug delivery system. In this review, we first anatomically and molecularly define the bbb and the underlying mechanisms of bbb transport under physiological conditions. we then explore changes and dysfunction of the bbb associated with normal ageing, neurodegeneration and acute brain injuries. We describe novel strategies to enhance penetration of the blood–brain barrier focused on those that have been developed over the past 5 years and have advanced into clinical trials. the order of strategies presented is from the oldest first to the newest last.
