Fluoropyrimidines Tipsheet
Introducing Tip Sheets Youtube The following information is intended for use by providers who are attempting to prescribe fluoropyrimidines, including fluorouracil (5 fu) and capecitabine to patients with certain dpyd genetic variants. Fluoropyrimidines (fp), a class of antineoplastic antimetabolites, first described in 1957 (heidelberger et al., 1957), have been a foundational component of chemotherapy regimens to treat a wide range of gastrointestinal (gi) malignancies for over half a century.
Capecitabine And Other Oral Fluoropyrimidines Youtube The fluoropyrimidines are broken down into three metabolites that have pharmacodynamic effects, fdump, fluoro deoxyuridine triphosphate (fdutp), and fluorouridine triphosphate (futp) (see pharmacokinetics pathway for more details) that act through these different mechanisms. Clinical champions who prescribe fluoropyrimidines can provide key insights into institutional standard of care practices and clinical workflows, while also encouraging the adoption of dpyd testing among colleagues. Fluoropyrimidines (5 fluorouracil, capecitabine and the oral prodrug tegafur) are used as the basis of adjuvant and palliative treatment for colorectal, oesophago gastric, breast and head and neck cancers. Fluoropyrimidines, including capecitabine and 5 fluorouracil (5 fu), remain the foundation of most chemotherapy regimens for gastrointestinal cancers. 1 these agents are commonly used either as monotherapy or in combination with other agents such as oxaliplatin and or irinotecan.

Oral fluoropyrimidines, including capecitabine and uft (uracil fluorouracil), have emerged as critical components in the treatment of cancer and fungal infections. In this review we describe the current knowledge on pharmacogenomics of fp related genes, beyond dpyd, focusing on fp toxicity risk and genetic effects on fp dose reductions. Model non tissue specific cancer cell displaying genes which may be involved in the pharmacodynamics of the fluoropyrimidines, 5 fluorouracil (5 fu), capecitabine and tegafur. pathway images and data are available under a creative commons by sa 4.0 license. We review methods for 5 fu synthesis, including the incorporation of radioactive and stable isotopes to study 5 fu metabolism and biodistribution. we also review methods for preparing rna and dna substituted with fps for biophysical and mechanistic studies.
