Honestly, the second run is tougher for a variety of reasons, so if you just want to purely enjoy the story, play claire 1st run. you'll get the same side of her story. the only difference is you don't see the final boss hinted at at the end of the first run (in the train). It's a bit confusing, because on the 2nd run you see minor evidence that the 1st run character was around, but it's to be assumed that they went through by unknown ways. so in the end, i suppose the "best" scenario is b but just because it lets you see the ending.
